What it is

How PT-141 works

PT-141, also called bremelanotide, acts on melanocortin receptors in the brain, part of the system involved in sexual desire and arousal.

In the brainDesire and arousal

Works on signaling in the brain rather than on blood flow.

Different fromSildenafil and tadalafil

Those medications act on blood flow. PT-141 acts in the brain.

An injectable form of bremelanotide, Vyleesi, is FDA approved for premenopausal women with acquired, generalized hypoactive sexual desire disorder (HSDD). Compounded PT-141 is not Vyleesi and is not FDA approved.

How you use it

PT-141 is used as needed, not every day. Your provider sets your dose, and your pharmacy label tells you how to use the spray.

1dose in 24 hours, at most
8doses a month, at most

Limits from the Vyleesi label

  • Timing

Used ahead of sexual activity. In the Vyleesi trials, the dose was taken at least 45 minutes before.

  • First doses

Nausea is the most common side effect. Your provider can adjust your dose or plan if it gets in the way.

  • Fine-tuning

Your provider looks at how it's working and how you're feeling before changing anything.

  • How you talk it through

Message your provider in your patient portal, or book a video visit at no extra cost.

Limits and timing as described for Vyleesi (bremelanotide) injection. Your provider's directions and your pharmacy label come first.

The research

What the studies found

Bremelanotide was tested in two large trials as Vyleesi, an injection. Compounded PT-141 was not the drug studied in these trials.

Vyleesi compared with placebo

The RECONNECT trials

1,267premenopausal women with HSDD
24weeks of treatment

"VYLEESI improved sexual desire and decreased the level of feeling bothered by low sexual desire in comparison to placebo."

U.S. Food and Drug Administration, Drug Trials Snapshot: Vyleesi. Trials published by Kingsberg et al., Obstetrics & Gynecology, 2019.
The nasal spray

Why the approved version is an injection

Bremelanotide was first developed as a nasal spray. That program stopped after some trial participants had increases in blood pressure, and the developer moved to an injection. The developer said:

"We believe this was due to variability in drug uptake with intranasal administration."

Palatin Technologies, as reported by Medical News Today.

Results from Vyleesi trials do not predict results with compounded PT-141. Compounded medications are not FDA approved. Individual results vary.

Questions

PT-141 questions

Is PT-141 the same as Vyleesi?

No. Vyleesi is an FDA-approved bremelanotide injection. Compounded PT-141 is made by a state-licensed pharmacy for one patient, from a prescription. It is not FDA approved, and the FDA does not review compounded drugs for safety, effectiveness or quality.

Who is PT-141 for?

Adults looking for help with sexual desire or arousal, when a provider decides it's appropriate. Vyleesi's approval covers premenopausal women with HSDD. Prescribing it for anyone else is up to your provider.

Is it the same as Viagra or Cialis?

No. Sildenafil (Viagra) and tadalafil (Cialis) work on blood flow. PT-141 works in the brain, on desire and arousal.

Can I use it every day?

No. It's used as needed. The Vyleesi label limits use to one dose in 24 hours and eight doses a month.

How is it shipped?

Do I need insurance?

No. Insurance isn't needed, and your visit, online or on video, is included in the treatment price.
